Listing a study does not mean it has been evaluated by the U. Treatment of GBM xenografts in vivo with chloroquine (CQ), an antimalarial agent, has been shown to reduce the hypoxic fraction and sensitizes tumors to radiation. Patients with a glioblastoma (GBM) have a poor prognosis with a median survival of 14.6 months after maximal treatment with a resection and chemoradiation.

I did the Stupp protocol exactly as you described -- radiation with Temodar, one month off, then six months of 5/23 Temodar. I was told that with this treatment I likely would have 12 to 18 months, and the average was 15 months overall survival, with about a 99% likelihood of recurrence during the first 12 months. Autophagy inhibitor chloroquine has shown moderate efficacy in a phase III clinical trial;11 meanwhile, the combination of thalido-mide and TMZ has been studied in phase II trials with promising results.12,13 We hypothesized that the combination of oncolytic adeno-virus and low-dose pulse of TMZ can lead to improved efficacy The results of RTOG 9802 make PCV appear to be a reasonable option, but RTOG 0424 Temodar results are just as good. My doctor's say Temodar will be as effective as PCV and more tolerable. The advantages of PCV are the RTOG 9802 survival plot for IDH1 mutations looks good, it's a multi-agent treatment, the methylation status isn't a factor, and there seems to be less risk of hypermutation.

The most common EGFR mutation in GBM (EGFRv III) is present in 50-60% of patients whose tumor shows amplification of EGFR. Epidermal growth factor receptor (EGFR) amplification or mutation is regularly observed GBM and is thought to be a major contributor to radioresistance.

Chloroquine is the generic form of the brand-name prescription medicine Aralen, which is used to prevent and treat malaria — a mosquito-borne disease caused by a parasite — and to treat amebiasis, an infection of the intestines caused by a parasite. Three cohorts of 3 patients will receive chloroquine in escalating doses 3 dose levels 200 mg up to 600 mg daily during standard treatment radiotherapy and temozolomide for newly diagnosed GBM. Extra patients can be added to a cohort in case of dose limiting toxicity, resulting in a maximum of 6 patients per dose level. Chloroquine is a medication used to prevent and to treat malaria in areas where malaria is known to be sensitive to its effects. Certain types of malaria, resistant strains, and complicated cases typically require different or additional medication.

Chloroquine has long been used in the treatment or prevention of malaria from Plasmodium vivax, P. malariae, excluding the malaria parasite Plasmodium falciparum, for it started to develop widespread resistance to it. Been told I have high liver enzymes. I have had very high liver enzymes that range from slightly elevated to over 400 for 1 of them, but I'm not sure which one. I have also had Hepatitis with a severe Lupus flare. My Gastroenterologist said the elevated liver enzymes could just be from fatty liver, but my Rheumatologist doesn't think so, he thinks it is Lupus related.
