If you use 6-well or 12-well plates, total volume of the medium should be 3ml and 1.5ml, respectively, and decrease the amount of each reagent accordingly.2. One hour prior to the transfection, change to medium containing 25M chloroquine (from x1000 stock in PBS, stored at -20C). (Chloroquine can be omitted, but increases efficiency about x2) to a final volume of 500 l.

Chemical transfection methods typically rely on electrostatic interactions to bind nucleic acid and to target cell membranes. These methods utilize compounds such as calcium phosphate, polycations e.g. DEAE-dextran, and liposomes, as well as more current technologies such as cationic lipids, polymers, dendrimers, and nanoparticles, to name a. Add 22 l of 25mM chloroquine to each 15cm dish for 10cm plate 7.8 l of chloroquinei.e. final concentration = 25 M. The cells should be transfected within 5-10’ of adding the chloroquine. 7. For 15cm plates, add 1.69ml of the transfection buffer to the DNA-CaCl2 solution by “bubbling”. Protocol for Lentivial Vector LV Production 2nd Generation Packaging Lenti virus can be packaged by any transfection reagent. In this protocol, we describe two methods a calcium phosphate precipitation method and an iMFectin Poly DNA method.

Add this mixture directly to the cells dropwise through the medium. As long as the cells are alive, they produce proteins. When you are ready, add 500 l of 2x HBS dropwise while gently mixing. Make sure you evenly sprinkle the droplet over the entire area.5. For secreted protein, you can change media once (at day 3 or 4, save the collected sup) and give it another 3-4 days of culture.

Chloroquine phosphate tablets, Chloroquine phosphate, USP, is a 4-aminoquinoline compound for oral administration. It is a white, odorless, bitter tasting, crystalline substance, freely soluble inwater. Chloroquine phosphate tablets are an antimalarial and amebicidal drug. Chloroquine is an inhibitor of the lysosomal degradation of the DNA which is taken up by the cells, so as leelee said, transfection should have been successful, albeit at a slightly lower level than if you would have added the chloroquine. In my normal transfections, I never add chloroquine and get high efficiencies nevertheless. Calcium phosphate transfection is the method of choice to produce long-term stable transfectants. This method also works well for transient expression of transfected genes and can be used with most adherent cell lines. 2. Plaquenil, diarrhea and weight loss? My husband has IBS and doesn't have weight loss or problems gaining weight. Considering my daughters have CD though, I suppose it is a good idea to go ahead with the scopes. If they don't find anything, and I try going off the Plaquenil, what alternatives are there?
