    Chloroquine pharmacokinetics

Tell your doctor and pharmacist about all of your drugs (prescription or OTC, natural products, vitamins) and health problems. You must check to make sure that it is safe for you to take chloroquine with all of your drugs and health problems.

Chloroquine combined with primaquine has been the standard radical curative regimen for Plasmodium vivax and Plasmodium ovale malaria for over half a century. In an open-label crossover pharmacokinetic study, 16 healthy volunteers 4 males and 12 females aged 20 to 47 years were randomized into two groups of three sequential hospital admissions to receive a single oral dose of 30 mg base primaquine, 600 mg base chloroquine, and the two drugs together. Chloroquine phosphate is a 4-aminoquinolone that acts by inhibiting heme polymerase. 198 Chloroquine is well tolerated and in appropriate doses may be safely used by young children and pregnant women. 199 Transient nausea or vomiting is seen in a small proportion of individuals and may usually be reduced by taking the drug with food. Occasionally, headaches or blurred vision are reported. Sep 01, 2019 Hydroxychloroquine, like chloroquine, is a weak base and may exert its effect by concentrating in the acid vesicles of the parasite and by inhibiting polymerization of heme. It can also inhibit certain enzymes by its interaction with DNA.

For all uses of chloroquine: WARNING/CAUTION: Even though it may be rare, some people may have very bad and sometimes deadly side effects when taking a drug. Do not start, stop, or change the dose of any drug without checking with your doctor.

  4. The N‐dealkylated metabolites of chloroquine are of pharmacological and toxicological importance. It may, therefore, be necessary to monitor their plasma concentrations in addition to that of chloroquine in patients on long‐term chloroquine therapy.

Recent reports have highlighted that chloroquine CQ is capable of inhibiting ZIKV endocytosis in brain cells. We applied pharmacokinetic modeling to develop a predictive model for CQ exposure to identify an optimal maternal/fetal dosing regimen to prevent ZIKV endocytosis in brain cells. Pharmacokinets of Chloroquine. Absorption It is well absorbed after oral administration. Distribution It is distributed in protein bound form and concentrated in melanin containing cells. Metabolism some drugs undergo metabolism in the liver. Excretion It is excreted mainly through urine. Oct 01, 2018 Chloroquine is deposited in the tissues in considerable amounts. In animals, from 200 to 700 times the plasma concentration may be found in the liver, spleen, kidney, and lung; leukocytes also concentrate the drug. The brain and spinal cord, in contrast, contain only 10 to 30 times the amount present in plasma.

Plaquenil and drinking - Sjogren's World I also take Methotrexate, along with Plaquenil plus many others and I don't drink any alcohol. Then again, this is an individual decision. I would run it by my doctor and see if they thought it was ok. You then could either drink a little wine, or abstain. I have read where a glass of wine OCCASIONALLY wouldn't hurt.
